Got an open port on a hydraulic manifold I need to cap off during a rebuild, this the kind of fitting that handles that?

Flat face blind flanges like this one are exactly what you want for that situation. The CD62 series from Anchor Fluid Power is built for port blocking and line termination, so capping off an unused or inactive port during assembly or maintenance is its primary job. Just confirm your open port uses a #16 SAE flange connection and you should be in good shape.
