what's the difference in size between the male and female ends on this adapter? trying to step up from a 3/8 line to a 1/2 connection and want to make sure I got the right part

The male end threads are 3/8-18 NPT and the female end is 1/2-14 NPT, so yes, this steps up from the smaller male side to the larger female side in a straight configuration. That's exactly the size combination you're describing.
