Will this fitting work with a standard Teflon hose?

This Eaton Aeroquip fitting is designed for use with a 1-wire braid Teflon hose.

Does this fitting need any kind of thread sealant or mandrel to put it together, or is it pretty much ready to go as-is?

No mandrel and no thread sealant required for installation. The reusable stainless steel construction and the JIC 37-degree flare swivel connection handle the seal on their own, so you're not adding extra steps or materials to the assembly process.
