Got a port on my hydraulic manifold threaded 9/16-18 ORB and I need to run 1/4 inch tubing off it with a JIC connection. this fitting gonna bridge that gap straight inline?

That's exactly what it does. One end threads into your 9/16-18 SAE O-Ring Boss port, and the other side is a 7/16-20 male JIC 37 degree flare, which is the standard connection for 1/4 inch OD tubing. The whole thing runs straight through at 1.29 inches overall, so there's no offset or angle to deal with. The ORB end seals against the port face with an o-ring, which keeps that connection tight without relying on thread fit alone.

i’ve got a 9/16-18 ORB port but my hose end is 1/4 tube JIC, is this the right little adapter to tie those together?

For a straight, inline hookup between those two standards, this adapter matches a 9/16-18 Male SAE O-Ring Boss on one side and a 7/16-20 Male JIC 37 degree flare sized for 1/4 inch OD tubing on the other.

Will this here fitting work with a regular 1/4 inch tube?

Yes sir, this adapter is made to work with 1/4" OD tubing, using the 7/16-20 Male JIC 37° Flare connection.
