What size tube does the male end of this reducer hook up to?

The male JIC end uses a 7/16-20 thread, which corresponds to a quarter-inch tube OD. So if you're stepping down to a 0.25-inch line on one side, that's the end you're connecting to.

i’ve got a 1/4 inch tube line with a 7/16-20 JIC male on it, will this reducer hook up to that?

For a 1/4 inch tube OD JIC connection, the male side on this adapter is 7/16-20 JIC 37 degree flare, so it matches that size. The other end is a female JIC 37 degree flare, meant to accept a standard JIC flare coming from the larger side you are reducing from.
