Will this fitting hold up to some harsh chemicals, or should I be looking at something else?

Both ports are female NPT on this thing, so what exactly goes in between them? Like do I need two male nipples to make it work in a line?

Yeah, you'd run a male-threaded nipple or fitting into each port. Since both ends are 1/4-18 female NPT, you're connecting two male-threaded components together at a 90-degree angle. It's real common in tight hydraulic connections runs where you need to change direction but don't have room to swing a standard elbow with male ends. The fitting itself is only 0.88 inches long overall, so it fits into pretty confined spaces without a lot of fuss.
