We're running this on a press application at 4,500 psi. Does it need a steel subplate at that pressure or will a standard one hold up?

Anything over 3,000 psi does require a steel subplate or manifold with this valve. At 4,500 psi you're well past that threshold, so a standard subplate wouldn't be the right call. The valve itself is rated up to 5,075 psi, so the valve body isn't the limiting factor there, just the mounting surface beneath it.

got a D03 subplate setup already, is this one gonna bolt up or am i gonna be drilling and re-hydraulic connections

Fit is based on the ISO 4401 Size 03, NFPA D03 port interface and a subplate or manifold mount style, so it’s meant to mount on industry standard D03 surfaces without custom drilling or re-hydraulic connections.

Can this valve handle different types of hydraulic fluids, or is it strictly for mineral oil?

This valve is rated to handle mineral oil within a fluid temperature range of -4°F to 158°F. It can also handle water-based fluids, but the temperature range shifts to +50°F to 129°F.
