got a -40 code 61 flange head, is this kit the right size and what all comes in it?

For a -40 Code 61 split flange setup with a 3.31 inch flange head diameter, this kit matches that size. It includes two flange halves, four Grade 8 hex bolts, four lock washers, and one 90 durometer Buna-N O-ring.

what fluids is that buna-n o-ring gonna hold up to, petroleum based or water based or both?

Buna-N handles both petroleum and water-based hydraulic fluids just fine. That said, if your system runs anything outside those two categories, or operates at temperatures pushing past 250 degrees F, you'd want a different seal material. The kit itself is rated down to negative 40 degrees F on the cold end, so typical outdoor or unheated shop conditions aren't a concern.
