Will this brass fitting work with half inch tubing going to 3/8 NPT threads?

Yes, it handles exactly that connection - 0.50 inch tube OD on one end with 3/8-18 male NPT threads on the other. The push-connect side gives you that quick tube connection while the threaded end screws into your 3/8 NPT female port.
