i got 1/4 inch nylon tube on hand. does this fitting take that and thread into a 1/8 npt port?

It’s made for 0.25 inch tube OD on the push to connect side and a 1/8-27 male NPT thread on the other side. It’s also called out for use with PT230 and TP160 nylon tubing.

Will this fitting hold up okay if I'm running air through it, like for shop air tools and such?

This Eaton Aeroquip adapter is made from brass with nickel plating, so it's designed to be durable for compressed air and other pneumatic systems. It's rated up to 250 psi, which should be plenty for most shop air setups.
