Got a 3/8 tube line coming off a brass fitting and need to tee into it with a 1/4 NPT male thread on the run side. this the right part for that?

that lines up exactly with what this fitting does. The run side is 1/4-18 male NPT, and both the run and branch on the flare side are 5/8-18 male SAE 45-degree. The tube OD it's built for is 0.38 inches, which is your 3/8 tube. If your existing line matches those thread specs, you're in good shape to tee off without any adapters stacked on top of each other.
