got a hose end that’s 1/2 JIC, will this adapter screw right on and then go into an S16 DIN tube port?

The JIC side is a 3/4 - 16 UNF male thread sized for 1/2 inch JIC, and the other side is an M24 x 1.5 male thread made to match an S16 DIN tube connection (DIN 2353S sealed). If those are the two ends you are mating to, the thread sizes and types line up.

fitting goes from JIC on one end to DIN tube on the other -- what sizes exactly are we talking on both sides?

On the JIC side you've got a 3/4-16 UNF male thread, which is your standard 1/2" JIC size. The other end is M24 x 1.5 male thread, sized for S16 DIN tube connections. Both ends are male, so keep that in mind when you're planning your port or hose end connections.
