Will this quick disconnect work with hydraulic fluid, or is it just for water?

Designed for hydraulics and fluid transfer applications, this Eaton Aeroquip quick disconnect uses Buna-N seals which are compatible with many hydraulic fluids. You can also review the fluid compatibility document linked on the product page for more details.

does this thing actually connect when there's already pressure in the line, or do you have to bleed it down first?

Connecting under live pressure is one of the main things this coupling is built for. It can handle up to 500 psi when mating the halves, so in most working hydraulic setups on dump trucks or wet line systems, you won't need to depressurize before plugging in. That saves real time when you're out in the field and can't easily bleed the system down.
