does this cap work if there's already pressure built up on the line, or do i have to bleed it down first?

Pressure in the line is not a problem here. The 5100 Series dust cap is built to connect against pressure up to 500 psi, so you can make the connection without bleeding the system down first. That's one of the main reasons folks reach for this cap in working hydraulic setups.
