This 45VQ series vane pump has a 50 ring size, whereas the 45VQ40 would have a 40 ring size. You'll want to confirm that the 50 ring size is appropriate for your application.

got a 45VQ50 that needs replacing on a wheel loader, this the one with the 14 tooth splined shaft and clockwise rotation?

Correct on both counts. The shaft is a 14 tooth SAE D spline and rotation is clockwise. It also runs a 2-bolt SAE C mounting flange with SAE 4-bolt flange ports, 3 inch inlet and 1-1/2 inch outlet. The inlet port sits 90 degrees counterclockwise from the outlet. If those details match what you're pulling off, this is a direct fit.
