got a 3/8 inch line and need to go from hose to a threaded port. is this the right kind of fitting?

what exactly is the hose barb side connecting to on this fitting, is it meant to go onto regular rubber hose or something more rigid?

Hose barb fittings like this one are built to push into flexible hose and hold it in place with the barbed ridges. Rubber hydraulic hose or similar flexible tubing is exactly what the barb end is made for. The female pipe end on the other side threads onto a standard pipe fitting or threaded port, so this piece bridges your flexible hose run over to a fixed threaded connection in the system.
