Will this work for hooking up a quarter-inch NPT fitting to a 9/16 JIC?

Yes sir, this adapter does exactly that. It's made to connect a 1/4-18 Male NPTF (that's the quarter-inch NPT you mentioned) to a 9/16-18 Male JIC SAE 37° Flare fitting.
