Does the offset port setup on this cylinder cause any headaches when fitting it into tighter hydraulic configurations, or does it actually make things easier?

Offset port placement tends to help more than it hurts in tight setups. Routing lines becomes a lot more manageable when the port isn't centered, especially in systems where straight-on access is blocked by nearby components. For complex hydraulic builds where line routing is already a puzzle, that offset geometry usually saves more space than it costs.

trying to tuck lines in a tight spot, what does “off-set port” mean on this cylinder?

Offset port means the hydraulic port is positioned off the main centerline of the cylinder. That layout is meant to make hose and fitting routing easier in cramped or complex hydraulic setups, while still supporting smooth flow through the system.
