got a 1/4 inch male pipe fitting on my manifold block, will this adapter thread right onto that, and what size does the swivel end open up to?

The male end on this fitting is a 1/4-18 NPTF thread, so yes, that matches up to a standard 1/4 inch pipe port. The swivel end is a 1/2-14 NPSM female, which gives you a half-inch connection on the hose side. That size jump between the two ends is worth keeping in mind when you're planning your line routing.
